Bispecific antibodies are an artificial protein that is composed of fragments of two different monoclonal antibodies and consequently binds to two different types of antigens.
Over the past decades, advances in bioengineering and expanded insight in tumor immunology have resulted in the emergence of novel bispecific antibodies constructs. Pre-clinical studies of various bsAb constructs have shown impressive results in terms of immune effector cell retargeting, target dependent activation and the induction of anti-tumor responses. The most widely used application of this approach is in cancer immunotherapy.
They are capable of bridging two different molecules. There are at least 30 different BsAbs (such as tandem bites) and related molecules in clinical development currently, two of which have been approved to be put onto market, catumaxomab (REMOVAB®) and blinatumomab (BLINCYTO®), which also suggests that bsAbs can be developed as promising therapeutic agents.
