Creative Peptides Announced Chemical and Biological Production of Disulfide-rich Backbone-cyclized Peptides

Creative Peptides, one of the world's leading peptide suppliers, announced the release of production of disulfide-rich backbone-cyclized peptides using both chemical and biological methods.

New York - July 31, 2018 - Creative Peptides, one of the world’s leading peptide suppliers, announced the release of production of disulfide-rich backbone-cyclized peptides using both chemical and biological methods.

Over the last decade and more, disulfide-rich cyclic peptides have attracted great interest in the development of peptide-based therapeutics. Having exceptional stability toward chemical, enzymatic, or thermal attack, they have exciting potentials as leads or frameworks in drug discovery. In addition, they are also used as scaffolds and bioactive epitopes can be grafted onto them to take full advantage of the favorable biophysical properties of disulfide-rich cyclic peptides.

“In spite of the huge potentials, however, the use of disulfide-rich cyclic peptides is faced with some synthetic challenges, mainly associated with construction of the circular backbone and formation of the correct disulfides,” said Dr. Robert Martinez, Head of Creative Peptides’ synthesis businesses, in a statement.

Against such backdrop, a new service is launched by Creative Peptides to assist researchers in synthesizing disulfide-rich cyclic peptides using chemical or biological method. Cyclic peptides consisting of approximately 15-40 amino acids can be synthesized with solid-phase peptide synthesis (SPPS) technique. Two steps are involved with this process: production of linear precursor and cyclization. “Though chemical method is most frequently used, in recent years, enzyme-mediated cyclization has become a new promising technology due to its safety, efficiency, and cost-effectiveness,” revealed Dr. Robert Martinez.

“At Creative Peptides, backbone-cyclized polypeptides can be produced using standard heterologous expression systems such as E. coli or using protein trans-splicing method,” further explained Dr. Robert Martinez.

Benefits of disulfide-rich cyclic peptides

In general, cyclization significantly stabilizes peptides. And this benefit is amplified when a cyclic backbone is combined with disulfide bonds, which brings about exceptional stability to peptides. Over the last two decades, peptides with these features have been the focus of studies.
