Primary cells, such as Natural Killer Cells and T kill cells, most closely represent the tissue of origin. They are taken directly from the tissue and processed to establish them under optimized culture conditions. Because they are derived from tissues and are not modified, they are more similar to the in vivo state and exhibit normal physiology. Therefore, they provide an excellent model system for studying the normal physiology and biochemistry of cells (eg, metabolic studies, aging, signaling studies) and the effects of drugs and toxic compounds on cells. Keep in mind that primary cells have a limited life span and stop dividing (or aging) after a certain number of cell divisions, and are more difficult to culture and maintain than continuous cell lines. The variability induced in primary cells obtained from donor and subculture practices is a major challenge for researchers studying cell signaling pathways. Researchers are more likely to screen cells for sensitivity to common stimuli before beginning signaling studies. Pre-screened primary cells can save valuable resources because they stimulate the activation of major signaling pathways.
The most prevalent primary cell types used for research are epithelial cells, fibroblasts, keratinocytes, melanocytes, endothelial cells, muscle cells, hematopoietic cells, and mesenchymal stem cells. Cultures are initially heterogeneous (representing a mixture of cell types present in the tissue) and can be maintained in vitro only for a limited period of time. The primary cells can be manipulated for an infinite number of subcultures by an in vitro process called transformation. Transformation can occur spontaneously or can be induced chemically or virally. When primary cultures undergo genetic transformation (providing appropriate fresh media and space), they divide indefinitely and become immortalized secondary cell lines.
The use of primary cells avoids many of the moral objections raised against animal experiments. Experiments on human tissue are allowed, otherwise, these experiments cannot be performed in vivo. Meanwhile, the use of primary cells provides more relevant results than cell lines. Pre-screened primary cells are a very good model that closely represents signaling in vivo. More importantly, primary cells including Peripheral Blood MONOCYTES are cost effective because they help reduce the cost of animal models needed for in vivo research.
