Bispecific antibodies (BsAbs) recognize two different epitopes, based on the coupling of two antigen-recognizing elements into a single construct, which have reached outstanding clinical results. To develop BsAbs for treating diseases, it is critically important to perform comprehensive BsAb analysis to ensure their quality and function.
1. Biochemical characterization
Biochemical characterization plays an important role in the BsAb development. There are mainly three stages in biochemical characterization as follows:
- Purity Measurement: the sodium-dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) analysis, the liquid chromatography-mass spectrometry (LC-MS) analysis and partial-filling affinity capillary electrophoresis (PFACE) analysis.
- Molecular Weight/Mass Measurement: size-exclusion chromatography multi-angle light scattering (SEC-MALS), electrospray ionization mass spectrometry (ESI-MS) and electrospray ionization time-of-flight (ESI-TOF).
- Molecular Structure Analysis: nuclear magnetic resonance (NMR) spectroscopy, small-angle X-ray scattering (SAXS), and molecular docking analysis.
2. Functional analysis

3. Pharmacokinetic and pharmacodynamic (PK/PD) analysis
