The basic or primary structure of GFP is a 238 amino acid chain, weighing about 27,000 atomic mass units, and only about 4 amino acids directly produce any fluorescent effect. The secondary structure is a series of spirals and pleats caused by hydrogen bonds in the chain, while the tertiary structure is a barrel made of 11 plates, covered with a spiral. There is a chromophore located in the center, which is an amino acid responsible for the short-chain changes in luminescence. The barrel structure keeps the chromophore away from the solvent, allowing GFP to fluoresce under any condition and denaturation of proteins by heat and pH.
Biologists utilize GFP as a marker protein to study embryonic and fetal cells during development, as it can attach and label another fluorescent protein, enabling scientists to observe the presence of specific proteins in organic structures. With DNA recombination technology, scientists combine the GFP gene with another gene that produces the protein they want to study and then insert the complex into the cell. It is postulated that the cell expresses the target gene if the cell produces green fluorescence. In addition, GFP is utilized to label specific organelles, cells, and tissues. Due to the GFP gene is heritable, the progeny of the marker entity also exhibit green fluorescence.
