The chromosomal aberration test with cultured mammalian cells is one of the sensitive methods for predicting environmental mutagens and/or carcinogens, which is also a supplemental test for the Salmonella/microsome test (Ames test). Most chemical substances cause chromatid aberrations, some compounds induce only destructive aberrations at relatively high dose levels, while others induce more exchange-type aberrations at relatively lower dose levels.

Over the past 20 years, there have been considerable evolutionary changes in the in vitro detection of chromosomal aberrations (CA). Processing and sampling time has always been the focus of attention because we are trying to develop a program to detect weak genotoxins. Different approaches have emerged in different parts of the world, leading to the need for coordination. Meanwhile, we are increasingly questioning the conditions under which germs respond positively, and several situations have been described in which the fragmentation response is considered not biologically relevant. Therefore, it is correct to carefully consider whether the type of damage in the CA test is related to human health.

For the methods which can be applied for chromosomal aberration, currently, Creative Bioarray follows OECD guideline 4731 to conduct an in vitro mammalian chromosome aberration test. The proliferating mammalian cell cultures are exposed to test chemicals with and without a metabolically-activated exogenous source. After the test articles are exposed for an appropriate period of time, the cells are treated with mid-term stagnant substances and harvested. Chromosomal diffusion slides are then prepared and stained. The chromosomal aberrations of metaphase cells were analyzed under a microscope. Each study included negative vector control and positive controls.

At present, the available types of genetic testing include:

• Newborn screening, which can be used to identify genetic diseases that can be treated early in life after birth

• Diagnostic testing, which can be performed before birth or at any time during a person's life, but not for all genes or all genetic disorders, applying to identify or rule out specific genetic or chromosomal disorders

• Prenatal testing for the detection of prenatal fetal gene or chromosome changes

• Carrier test, being used to identify people carrying a copy of a gene mutation
